In the context of the complete ban on feed antibiotics, diarrhea prevention and control has become the focus of feeding management of weaned piglets. However, the causes of diarrhea in piglets are complex and diverse, and the relevant molecular mechanisms have yet to be analyzed.
The reporter learned from the Institute of Subtropical Agroecology of the Chinese Academy of Sciences on June 28 that the team of Yin Yulong, an academician of the Chinese Academy of Engineering and chief researcher of the Institute, found that the composition of intestinal microorganisms, metabolites and miRNAs in diarrhea piglets caused by weaning stress has changed significantly. Among them, the deletion of ssc-miRNA-425-5p and ssc-miRNA-423-3p led to an increase in the abundance of Prevotella spp. and an increase in its fumarate reductase expression, so the resulting succinic acid was enriched in the intestine.
The team found that single-cell sequencing suggested impaired function and enhanced immune response of colonic epithelial cells in diarrhea piglets. Researchers used in vitro experiments to further explore the relevant mechanisms and found that on the one hand, the enriched succinic acid led to increased secretion of intestinal epithelial cells by regulating chloride ion secretion; Succinic acid, on the other hand, promotes the inflammatory response by activating the MyD88-dependent TLR4 signaling pathway.
This study suggests that decreased expression of miRNAs leads to the enrichment of succinic acid produced by microorganisms in the intestine, and excessive succinic acid promotes intestinal secretion and inflammatory response, resulting in diarrhea; It further enriches the occurrence mechanism of piglet diarrhea and provides a theoretical reference for diarrhea prevention and control.
